چکیده

Abstract Memory B cell is a heterogenous population and such heterogeneity focus of active investigation. How distinct subsets memory cells respond to repeated novel antigen challenges as SARS-CoV-2 in human remains subject vigorous research. We employed fluorochrome-conjugated streptavidin tetramers bound with RBD wild-type omicron variant (B.1.1.529), respectively, sort RBD-specific performed CITE-seq investigate detailed dynamics transcriptomes, surface phenotyes clonality healthy mRNA vaccination cohort. PBMC samples 8 BNT162b2 vaccine recipients from 7 consecutive timepoints, before after each dose 3-times vaccination, were analyzed. found pulsating, yet increasing trend, frequency atypical among tetramer-binding vaccine. SCENIC analysis find subset-specific regulatory gene networks revealed have regulons genes related activation receptor clustering, signaling calcium-dependent compared classical only one shared clone between cells, suggesting infrequent interconversion the two subsets. Furthermore, in-vitro polyclonal stimulation showed higher antibody-secreting (ASC) pre-plasmablasts flow cytometry ASCs IgG ELISPOT assay, cells. These findings highlight effector memory-like features viral challenge potential target future design.

Malaria-associated atypical memory B cells exhibit markedly reduced B cell receptor signaling and effector function

Protective antibodies in Plasmodium falciparum malaria are only acquired after years of repeated infections. Chronic malaria exposure is associated with a large increase in atypical memory B cells (MBCs) that resemble B cells expanded in a variety of persistent viral infections. Understanding the function of atypical MBCs and their relationship to classical MBCs will be critical to developing e...

Analysis of Sars-CoV-2 RBD Mutations in Khuzestan Province, Iran—A Retrospective Study, 2021

Background and Objective: One of the main reasons for the ongoing pandemic is the substitutions in the Receptor binding domain(RBD), which is involved in binding to the ACE2 receptor. Methods: Oro-nasopharyngeal swabs were collected from 41 confirmed patients between September 2020 and May 2021 to analyze the mutations of SARS-CoV-2 RBD in Khuzestan Province. Bi-Directional sequencing was use...

Tolerogenic Vaccination Reduced Effector Memory CD4 T Cells and Induced Effector Memory Treg Cells for Type I Diabetes Treatment

BACKGROUND Vaccination could induce immune tolerance and protected NOD mice from the development of type I diabetes (T1D). We previously demonstrated that insulin peptide (B9-23) combined with dexamethasone (DEX) stimulated the expansion of antigen specific regulatory T (Treg) cells which in turn effectively prevented T1D in NOD mice. Here, we aimed to investigate the therapeutic effect of tole...

Virus-specific memory CD8 T cells provide substantial protection from lethal severe acute respiratory syndrome coronavirus infection.

UNLABELLED Severe acute respiratory syndrome coronavirus (SARS-CoV) caused an acute human respiratory illness with high morbidity and mortality in 2002-2003. Several studies have demonstrated the role of neutralizing antibodies induced by the spike (S) glycoprotein in protecting susceptible hosts from lethal infection. However, the anti-SARS-CoV antibody response is short-lived in patients who ...
